چکیده

Digital holographic microscopy (DHM) is a modern technology allowing three-dimensional (3D) real-time non-destructive marker-free quantitative-phase imaging of inspected samples. DHM ranges from biological and biomedical applications to engineered surfaces metrology. In this Chapter we review the fundamentals of DHM. Starting from a theoretical analysis, we overview the main application fields of DHM. This Chapter introduces the reader in the field of DHM while providing a complete bibliography for a deeper study. As an example of application, we present the analysis of the capabilities provided by DHM when studying a fixed swine sperm biosample.

Principles and applications

In digital holography holograms are recorded by a CCD and image reconstruction is performed by a computer. It is free from tedious photographic processing and delivers three-dimensional distributions of both amplitude and phase quantitatively. Its main limitation that is caused by much lower resolution of CCDs than photographic materials has been substantially overcome by phase-shifting digital...

Measuring the deposited energy from a non-ionizing laser beam in water by digital holographic interferometry

Digital Holographic interferometry is a powerful and widely used optical technique for accurate measurement of variations in physical quantities such as density, refractive index, and etc. In this study, an experimental digital holographic interferometry setup was designed and used to measure the amount of energy changes induced by absorption of radiation from a non-ionizing infrared laser beam...

Glucose-Sensitive Holographic (Bio)Sensors: Fundamentals and Applications

Nowadays sensors and especially biosensors play an important role in medical diagnosis and detection of food and environment contaminations. Biosensors’ facilities have been improved significantly by using technologies such as surface plasmon resonance, microfluidics, laser, and electrochemistry. These technologies are now available on chips in micro- and nano-scale and are capable of mea...

Simultaneous two-color imaging in digital holographic microscopy

We demonstrate the use of two-color digital holographic microscopy (DHM) for imaging microbiological subjects. The use of two wavelengths significantly reduces artifacts present in the reconstructed data, allowing us to image weakly-scattering objects in close proximity to strongly-scattering objects. We demonstrate this by reconstructing the shape of the flagellum of a unicellular eukaryotic p...

Lensless digital holographic microscopy and its applications in biomedicine and environmental monitoring.

Optical compound microscope has been a major tool in biomedical imaging for centuries. Its performance relies on relatively complicated, bulky and expensive lenses and alignment mechanics. In contrast, the lensless microscope digitally reconstructs microscopic images of specimens without using any lenses, as a result of which it can be made much smaller, lighter and lower-cost. Furthermore, the...
